NEW ACTIVE PACLITAXEL DERIVATIVES' WITH IMPROVED WATER SOLUBILITY.
Renée Paradis* and Michel Pagé, Department of Biochemistry, Faculty of Medicine, Laval University, Ste-Foy, Quebec, Gl K 7P4
New paclitaxel amino acid derivatives substituted at the 2' position of the side chain were prepared. The first step consisted in introducing a glutaryl group at the 2' position by esterification of the free hydroxyl followed by the reaction of a peptide link between the carboxyl of the glutaryl portion and the amino terminal group of the amino acid. The derivatives were cytotoxic in vitro against many sensitive cell lines. They also increased G2+M phase arrest. Moreover, those derivatives were stable for several months and showed a better solubility in water than the parent compound. It seems also that they are hydrolyzed at pH 4 which let us believe that they are lysed inside the cell to free paclitaxel which binds to microtubules. Research funded by Canadian Institute for Health & Research.
